Metabolomics

Spatial metabolomics, lipidomics, and N-glycomics: goals, protocols, and metadata standards.

KPMP Metabolomics technologies

Spatial metabolomics

Goal: Localize the small metabolite markers in tissue sections from human kidneys.

MALDI-MSI workflow for fresh frozen tissue, from cryosectioning to METASPACE annotation, beside metabolite images overlaid on a PAS image. A comparison shows glomeruli identified in autofluorescence, molecular ion, overlay and PAS images.

Protocol(s)

‍

Spatial lipidomics

Goal: Localize the lipid markers in tissue sections from human  kidneys.

MALDI-MSI workflow for fresh frozen tissue with lipid images: PC (36:1) enriched in glomeruli, PC (36:4) in non-glomerular tissue, and their overlay. The glomerular marker SM (34:1), m/z 725.5568, highlights glomeruli in the METASPACE output.

Spatial N-glycomics

Goal: Localize the N-glycan markers in tissue sections from human  kidneys

N-glycan imaging workflow for FFPE tissue: sectioning, antigen retrieval, PNGase F spraying, matrix application and MALDI spatial profiling, exported to METASPACE. A second panel maps the N-glycan Man5 across the tissue at 50 µm resolution.
